治療目的によって服用方法が異なります。更年期障害の治療（ホルモン補充療法：HRT）には、子宮がある方は一般的にプロゲステロン（黄体ホルモン）製剤との併用します。必ず医師の指示に従って正しく服用してください。
通常成人は、1日0.625〜1.25mgを服用します。機能性子宮出血・腟炎に対しては、1日0.625〜3.75mgを服用します。症状や年齢に応じて適宜増減します。
血液が凝固しやすくなる血栓症（四肢、肺、心、脳、網膜等）の発症リスク、息切れ、胸の痛み、中枢神経症状（めまい、意識障害、四肢麻痺等）、下肢の疼痛や浮腫、急な呼吸困難、急性視力障害などがあります。
